SP 1

Systems Medicine of Heart Failure

The clinical proof-of-concept trial of the SMART study is organized as follows:
Within 6 subprojects the SMART study is planned to be realized. 60 patients with left ventricular pressure overload hypertrophy due to aortic valve stenosis (AS) (with or without Angiotensin receptor blockers are examined before and after Aortic Valve Replacement.

The figure provides a visual understanding of the planned proof-of-concept trial.

Examinations include pre- and postoperative parameters like clinical patient characteristics, medical treatment, labaratory parameters, cardiac magnetic resonance imaging data and transcriptomic and proteomic data from patient specific myocardial biopsies. These parameters are used to be applied to multilevel modelling tools for simulating and predicting of (pathophysiological) disease behaviour and evolution in response to surgical (Aortic Valve Replacement) and medical (Angiotensin receptor blockers) treatment. Validation of the simulated parameters is performed by comparing simulation based results to real outcome parameters of the individual patient.

Illustration of the SPs demonstrates the strong interactions between the different modelling methods.
