Development and functional characterization of pNET model systems
The main focus of the subproject lies in the establishment and biological characterization of pNET specific cell culture models as well as experimentally modified derivatives in vitro and in vivo. These cell culture models will be the main resources for the mathematical modelling of MAPK and mTOR signaling pathways. Protocols for the treatement with prototypic inhibitors for both signaling pathways will be established, and cell culture material for biochemical analysis of therapy-induced changes in signaling pathway activity will be produced. In cases where modeling and experimental validation of signaling pathway activity/alteration are consistent in terms of therapy response within a cell culture model, hypotheses will be tested in vivo in pNET Xenografts. In addition, the subproject will prepare and cultivate primary human NET cells, for the biochemical analysis of signaling components.
